What is Daurismo?

Daurismo (glasdegib) is used in combination with cytarabineto treat acute myeloid leukemia (AML).

Daurismo and cytarabine treatment is for use in adults who are 75 years or older, or who cannot use standard chemotherapy due to other medical conditions.

Before taking this medicine

Daurismo can cause severe birth defects or death of a baby if the mother or the father is taking this medicine at the time of conception or during pregnancy.

  • If you are a woman who is able to get pregnant, you may need to have a negative pregnancy test before starting this treatment. Use effective birth control to prevent pregnancy while you are using this medicine and for at least 30 days after your last dose.
  • If you are a man, use condoms when having sex with a woman who is able to get pregnant, even if you have had a vasectomy. Keep using condoms for at least 30 days after your last dose.
  • Tell your doctor right away if a pregnancy occurs while either the mother or the father is using Daurismo.

This medicine may affect fertility (ability to have children) in men. However, it is important to use birth control to prevent pregnancy because glasdegib can harm an unborn baby.

Do not breast-feed while using Daurismo, and for at least 30 days after your last dose.

To make sure Daurismo is safe for you, tell your doctor if you have ever had:

  • heart problems;
  • long QT syndrome (in you or a family member); or
  • an electrolyte imbalance (such as low levels of potassium or magnesium in your blood).

How should I take Daurismo?

Take Daurismo exactly as prescribed by your doctor, Follow all directions on your prescription label and read all medication guides or instruction sheets.

Daurismo is given in a 28-day treatment cycle, usually for a total of 6 cycles. Daurismo is taken by mouth, and cytarabine is given as an injection under your skin. Your doctor will determine how long to treat you with this medicine.

Take the medicine at the same time each day, with or without food.

Swallow the tablet whole and do not crush, chew, or break it.

You will need frequent medical tests. Your heart function may need to be checked using an electrocardiograph or ECG (sometimes called an EKG).

Store at room temperature away from moisture and heat.

Daurismo dosing information

Usual Adult Dose for Acute Myeloid Leukemia:

100 mg orally once a day on days 1 to 28 in combination with cytarabine 20 mg subcutaneously 2 times a day on days 1 to 10 of each 28-day cycle in the absence of unacceptable toxicity or loss of disease control; for patients without unacceptable toxicity, treat for a minimum of 6 cycles to allow time for clinical response

Usual Geriatric Dose for Acute Myeloid Leukemia:

What happens if I miss a dose?

Take the medicine as soon as you can, but skip the missed dose if your next dose is due in less than 12 hours. Do not use two doses at one time or use more than one dose in a 12-hour period.

What happens if I overdose?

Seek emergency medical attention or call the Poison Help line at 1-800-222-1222.

Overdose symptoms may include nauseavomitingdizziness, thirst, and little or no urination.

What should I avoid while taking Daurismo?

You must not donate blood or sperm while you are using Daurismo, and for at least 30 days after your last dose. If donated blood containing Daurismo is given to a pregnant woman, it could cause birth defects.

Daurismo side effects

Get emergency medical help if you have signs of an allergic reaction to Daurismo: hives; difficult breathing; swelling of your face, lips, tongue, or throat.

Call your doctor at once if you have:

  • fast or pounding heartbeats, fluttering in your chest, shortness of breath, and sudden dizziness (like you might pass out);
  • sores or white patches in or around your mouth, trouble swallowing or talking, dry mouth, bad breath, altered sense of taste;
  • cough with mucus, chest pain, feeling short of breath;
  • easy bruising or bleeding (nosebleeds, bleeding gums), purple or red spots under your skin;
  • red or pink urine, rectal bleeding, bloody or tarry stools;
  • abnormal vaginal bleeding;
  • cough with bloody mucus or vomit that looks like coffee grounds;
  • redness in or around your eyes;
  • bleeding from a skin wound, needle injection, or surgical incision;
  • any bleeding that will not stop;
  • low red blood cells (anemia) – pale skin, unusual tiredness, feeling light-headed, cold hands and feet; or
  • low white blood cell counts – fever, chills, mouth sores, skin sores, sore throat, cough, trouble breathing.

Your cancer treatments may be delayed or permanently discontinued if you have certain side effects.

Common Daurismo side effects may include:

  • bleeding;
  • low blood cell counts;
  • mouth sores, changes in your sense of taste;
  • loss of appetite, constipation;
  • rash;
  • muscle pain;
  • swelling in your arms or legs;
  • feeling tired; or
  • trouble breathing.

This is not a complete list of side effects and others may occur. Call your doctor for medical advice about side effects. You may report side effects to FDA at 1-800-FDA-1088.

What other drugs will affect Daurismo?

Sometimes it is not safe to use certain medications at the same time. Some drugs can affect your blood levels of other drugs you take, which may increase side effects or make the medications less effective.

Daurismo can cause a serious heart problem. Your risk may be higher if you also use certain other medicines for infections, asthma, heart problems, high blood pressuredepression, mental illness, cancer, malaria, or HIV.

Other drugs may interact with glasdegib, including prescription and over-the-counter medicines, vitamins, and herbal products. Tell your doctor about all your current medicines and any medicine you start or stop using.

Follow the following prescription medicine precautions :

  Daurismo may be harmful for your baby during pregnancy. Take proper medical help in case you are pregnant, may be pregnant or planning to become pregnant in the near future.
  This medicine is not indicated for use in women. It is not known if Daurismo is excreted in human milk. Because of the potential for serious adverse reactions in nursing infants from Daurismo a decision should be made to either discontinue nursing, or discontinue the drug taking into account the importance of the drug to the mother.
  Your health care professional can check blood tests to regularly monitor your liver and kidney function, while you are taking Daurismo . It is also important to check your levels of blood cells and other substances in the blood.
  Before taking this medicine, tell your doctor and pharmacist if you are allergic to Daurismo , any other medications, or any of the ingredients in Daurismo medicine. This medication is not recommended for use in patients with a known allergy to Daurismo .
  If you take Daurismo, you must stop drinking alcohol. This is one of the ways to prevent this disease from getting worse. If you are dependent on alcohol and want to stop drinking, your doctor can recommend a therapy that’s tailored for your needs.
  If you use other prescription drugs or over the counter products at the same time, the effects of this cancer medicine may change. Tell your doctor about all the drugs, vitamins, and herbal supplements you are using, so that your pharmacist can help you prevent or manage drug interactions.
  Missing a dose of Daurismo can upset your treatment plan. If you do miss one of your doses, take it as soon as you remember, unless it’s almost time to take your next dose. In this case, you should just skip the missed dose. Don’t take a double dose to “make up” for the one you missed. If you are not sure what to do after missing a dose, contact your doctor or pharmacist for advice.
  Do not take Daurismo the use by (expiry) date printed on the pack. Do not take it if the packaging is torn or shows signs of tampering. If it has expired or is damaged, return it to your pharmacist for disposal.
  During your treatment with Daurismo , it is very important for your doctor to know about your medical history if you have Kidney problems, hepatitis, liver problems, HIV (human immunodeficiency virus), triglycerides, diabetes or other health problems.

 

What are the storage instructions for Daurismo ?

Patients must properly store the drug to retain its quality and efficacy. Following storage instructions will be helpful in storing the drug in the best of manner.

  • Store Daurismo at room temperature of 25°C (77°F); excursions permitted to 15°C to 30°C (59°F to 86°F). Make sure to always keep your medicine in an area of your home that is out of the reach of children and pets.
